چکیده

Symbioses between nitrogen-fixing rhizobial bacteria and legumes are of great agricultural importance. We have recently-discovered a 15-amino acid peptide-hormone that contributes to root-nodule formation, essential for rhizobium-legume symbiosis. Manual application of these peptide-hormones increases nodule-formation, as does elevating activity of the corresponding gene in Medicago truncatula. The structure/bio-activity relationship of these hormones has however, until now, remained enigmatic. We report structural characteristics essential for the activity of our hormone, dubbed “D1A”. D1A variants, with alterations in key amino-acids, were analysed via ultraviolet circular dichroism (CD), which generates structure-specific spectra. Spectra were compared to existing literature. Reconciliation of structural data with previously-obtained hormonal-activity data suggests a ‘hairpin’ shape to be biologically-relevant, potentially affecting receptor-binding. We speculate D1A possesses a flexible central sequence of amino acids, facilitating this hairpin structure. Structural stabilisation requires interactions between the distal, aromatic amino acids, with natural hormone-structure also requiring proline-hydroxylation. Mono-hydroxylation resulted in hairpin destabilisation, whilst non-hydroxylation resulted in elevated hairpin character.

In vitro study of drug-protein interaction using electronic absorption, fluorescence, and circular dichroism spectroscopy

In the near future, design of a new generation of drugs targeting proteins will be required. Considering the complex bond between the drug and protein, the structure and stability of the target protein should be considered. So far, a series of in vitro investigations have been conducted with the aim of predicting drug-biological medium interactions. In these studies, use of spectroscopic method...

In Silico and in Vitroinvestigations on cry4aand cry11atoxins of Bacillus thuringiensis var Israelensis

In the present study we attempted to correlate the structure and function of the cry11a (72 kDa) and cry4a (135 kDa) proteins of Bacillus thuringiensis var israelensis. Homology modeling and secondary structure predictions were done to locate most probable regions for finding helices or strands in these proteins. The JPRED (JPRED consensus secondary structure prediction server) secondary struct...

IN BRIEF Medicago truncatula CRE1 Cytokinin Receptor Regulates Nodulation and Lateral Root Development

The plant hormone cytokinin is implicated in the control of root architecture and development, including legume root nodulation. Gonzalez-Rizzo et al. (pages 2680–2693) identified a Medicago truncatula homolog of Arabidopsis, CYTOKININ RESPONSE1 (CRE1), which encodes a cytokinin receptor histidine kinase, and made use of RNA interference of Mt CRE1 to investigate the role of cytokinin in the no...

Purification and Characterization of an Antibacterial, Antifungal and Non Hemolytic Peptide from Rana Ridibunda

Amphibians have a large variety of antimicrobial peptides that serve as natural innate barriers limiting microbial infection or, in some instances, act as an integral component in response to inflammation or microbial infection. A novel peptide with antibacterial effects and without hemolytic activity was purified from skin secretions of Rana ridibunda by multisteps cation-exchange FPLC, revers...
